追赶生长大鼠肝组织瘦素受体的变化及其相关因素研究

摘    要:目的探讨追赶生长过程中大鼠肝组织瘦素受体的变化及其对葡萄糖代谢的影响。方法 5周龄雄性Wistar大鼠适应性喂养一周后随机分为对照组和追赶生长组。追赶生长组给予对照组半量饲料喂养4周后开放饮食8周;对照组则持续开放饮食喂养。于实验的0周、4周、6周、8周、12周采集血液及肝脏。用ELISA法测定血清瘦素水平;采用免疫组化法检测肝细胞表面瘦素受体表达,全自动生化检测仪检测血糖,放射免疫法测定胰岛素水平。结果实验第4周(半量喂养期),追赶生长组大鼠的血清瘦素水平明显低于对照组,随着开放饮食时间的延长,瘦素水平逐渐增加。在实验终点(第12周),追赶生长组肝组织表面瘦素受体水平低于对照组,而瘦素水平、血糖水平、胰岛素抵抗指数IRI明显高于对照组,差异均具有统计学意义(P均〈0.05);肝组织表面瘦素受体水平与血清瘦素及HOMA-IR呈显著负相关(P均〈0.05)。结论追赶生长可导致肝细胞表面瘦素受体水平降低,肝细胞表面瘦素瘦素受体下降与追赶组大鼠葡萄糖代谢异常有关。

Abstract:Objective To explore the expression of liver leptin receptor in catch-up growth rat and influence on glucose metabolism.Methods Five-week old male Wistar rats were fed free for one week,and randomly assigned into NC group and RN group,and RN group(refeeding normal chow group after food restriction) were fed with normal diet for another 8 weeks after semistarvation for 4 weeks,while NC group was given free food for 12 weeks.Blood and liver tissue were collected on the 0,4th,6th,8th,12th week respectively,the serum leptin was determined using ELISA method,the leptin receptor was detected using immunochemistry.Serum glucose was analyzed using automatic biochemistry detection machine.Serum insulin was detected using radioimmunoassay.Results On the 4th week,the serum leptin level of RN group was significantly lower than that of NC group,and it increased as the time went on.At the end of the experiment,liver leptin receptor was lower than that of NC group(P<0.05),while the serum leptin,serum glucose and HOMA-IR of RN group were significantly higher than those of NC group(all P<0.05).The expression of leptin receptor negatively related with the serum leptin,HOMAIR at the 12th week.Conclusion Catch-up growth could down regulate the expression of liver leptin receptor,which maybe related with the abnormality of the glucose metabolism.
