单克隆抗体MAb7及其相应抗原在单核－巨噬细胞系统及中性粒细胞中的表达

目的：建立一株抗人单核－巨噬细胞及中性粒细胞的杂交瘤ＭＡｂ７．方法：用人肝癌组织细胞悬液免疫ＢＡＬＢ／ｃ小鼠，取脾细胞与ＳＰ２／０细胞融合，ＡＢＣ染色法筛选单克隆抗体，ＡＰＡＡＰ法鉴定髓细胞特征。结果：抗体亚类为小鼠ＩｇＧｌ，相应抗原经亲和层析纯化，Ｍｒ６００００．免疫组化染色，在石蜡切片的组织中和在血片及骨髓涂片中单核巨噬细胞和成熟的中性粒细胞阳性，单核细胞性白血病及中性粒细胞性白血病阳性，而上皮、内皮、网状组织细胞、成纤维细胞、平滑肌细胞及其它粒细胞、淋巴细胞、红细胞均为阴性．结论：ＭＡｂ７在研究单核－巨噬细胞的分化、免疫功能及病理分型等方面有一定的意义和价值．

Monoclonal antibody MA67 and its related antigen expressed inmonocyte/macrophage lineage and neutrophils

objective :To establish a monoclonal antibody MAb 7.Metbods :The BALB/c mice was immu- nized with human hepatoma cell suspension.The spleen cells and the SP2/0 myeloma cells were fused.The monoclonal antibody was selected vvith ABC staining method and the character of marrow cells identified with APAAP method. Resulb:The subclass of MAb7 is murine IgGl.Its related antigen was purified by affinity chromatography whose molecule weight was 60000 daltons.Immunohistochemical(ABC, APAAP)staining showed that MAb7 reacted with macrophages of almost all known tissues,peripheral blood monocytes,neo- plastic macrophages (leukemia)and mature neutrophils, but not with epithelial and endothelial cells,fibrob- lasts,smooth muscle and reticulum cells,lymphocytes and erythrocytes.Conclusion :MAb7 is of definite value to pathologic diagnosis and the studies of monocyte/macrophage differentiation and immunologic func- tions.
