乙酰半胱氨酸拮抗三氧化二砷诱导肝癌细胞凋亡效应的初步研究

摘    要:目的:探讨抗氧化剂乙酰半胱氨酸(NAC)能否抑制三氧化二砷(AT)诱导肝癌细胞凋亡及其可能的机制。方法:采用四甲基偶氮唑蓝(MTT)、DNA梯形电泳及苏木精-伊红染色的方法观察AT对人肝癌细胞BEL-7402的杀伤效应、诱导凋亡效应以及NAC对该效应的影响。结果:①适量AT能抑制肝癌细胞的恶性生长,诱导细胞凋亡;②一定量的NAC能拮抗AT对肝癌细胞的杀伤效应及诱导凋亡效应。结论;AT对肝癌细胞的作用与细胞内谷胱甘肽有密切关系;很可能通过与细胞内巯基(-SH)结合,导致多种功能蛋白失活来诱导细胞凋亡,抑制肝癌细胞的恶性生长。

Abstract:Objectives To study the antagonising effect of antioxidant N-acetylcysteine (NAC) on arsenic trioxide (AT)-induced human hepatocarcinoma cell apoptosis and its possible mechanisms.?Methods MTT, DNA ladder and hematoxylin-eosin (HE) staining methods were used to study the effects of AT on the human hepatocarcinoma cell line BEL-7402 in inducing apoptosis and producing injuries in the cells,as well as the antagonistic action of NAC in vitro.?Results (1) AT in effective doses inhibited the proliferation of human hepatocarcinoma cells and induced apoptosis. (2) NAC could antagonise the apoptosis and cell injuries induced by AT.?Conclusions The effect of AT on cancer cells is related to the intracellular GSH level. AT might inactivate many functional proteins by binding with their -SH radicles, inhibiting the abnormal growth of human hepatocarcinoma cells and inducing their apoptosis.
