肺癌患者痰液脱落细胞端粒酶活性检测

目的　探讨痰液脱落细胞端粒酶活性表达作为肺癌早期无创伤性辅助诊断指标的可能性。方法　采用必嗽平注射液液化痰液收集脱落细胞 ,逆转录一聚合酶链反应一聚丙烯酰胺凝胶电泳 (RT -PCR -PAGE)进行脱落细胞端粒酶活性检测 ,同时采用巴氏染色法进行脱落细胞学检查。结果　 6 0例肺癌患者痰液脱落细胞中有 35例检测到端粒酶活性 ,阳性率为 5 8.3% ,而 2 0例良性支气管 -肺病变患者痰液脱落细胞内无一例检测到端粒酶活性 ,两者差异有显著性 (P <0 .0 5 ) ,6 0例肺癌患者痰液脱落细胞学检查明确为癌细胞者 2 9例 ,阳性率 4 8.3% ,与端粒酶活性阳性表达率之间差异无显著性 (P >0 .0 5 ) ,肺癌患者痰液脱落细胞端粒酶活性阳性表达与病变部位及组织学类型无相关性 (P >0 .0 5 )。结论　痰液脱落细胞端粒酶活性检测可作为肺癌早期无创伤性检查辅助诊断指标 ,端粒酶活性阳性表达率与肿瘤部位、组织病理学类型无关

Detection of Telomerase Activity in Exfoliated Cells in Sputa from Patients withLung carcinoma

Objective To investigate the expression of telomerase activity in exfoliated cells and determine whether its detection is of value in the early diagnosis of lung carcinoms.Methods The sputa from benign and malignant bronchal-pulmonary diseases were lisquated by Bromhexine Injection and exfoliated cells were collected,RT-PCR-PAGE was employed to detect the expression of telomerase activity,The standard Papanicolaou Staining was used for cytologic examination.Results 35samples had been detected telomerase activity in sputa from patients with lung carcinomas,none of the 20 patients with benign bronchial-pulmonary diseases was detected telomerase activity in exfoliated cells in sputa , which had a statistically significant difference(P<0.05),29 of the 60 samples were found cancer cells in sputa through cytological examination,there were no differences between cytological examination and the expression of sputa through cytological examination and the expression of telomerase acitivyty,The expression of telomerase activity was not related to the location and pathological classifications of lung carcinomas.Conclusions The telomerase activity in exfoliated cells in sputa may serve as a tumor marker for the early diagnosis of lung carcinomas.The expression of telmerase activity is irrelevant with the location and pathological classifications of lung carcinomas.
