复制型HBV转基因小鼠肝组织HBcAg分布特性分析

摘    要:目的探讨复制型HBV转基因小鼠肝组织HBcAg的分布特性。方法通过图像分析软件,对转基因小鼠的肝组织HBcAg在肝内不同区域以及肝细胞核内的表达与分布进行系统和量化分析。结果①HBcAg在复制型HBV转基因小鼠肝组织内高表达,弥散分布,主要为胞核型,少数为胞浆型;②在40倍物镜下,200万个像素单位内,肝组织HBcAg阳性细胞核平均个数92.85±50.15,阳性细胞核平均染色面积(70965.80±37919.20)像素单位,阳性细胞核平均光密度0.67±0.03,光密度比值2.37±0.09;③在肝细胞核内,HBcAg的表达不均匀,核周边的表达量较多。结论四项指标经统计分析,在门管区和中央静脉周围两个区间,HBcAg阳性细胞核平均个数、平均染色面积、平均光密度以及阳性细胞核与背景的光密度比值均没有统计学差异,且性别差异不显著。

Abstract:Objective To discuss the distribution features of HBcAg in replicative HBV transgenic mouse hepatic tissue. Methods The expression and distribution of HBcAg in different liver areas and the nucleus of HBV-transgenic mice were analyzed systematically and quantitatively by an image analysis software. Results (1) HBcAg was highly expressed in replicative HBV-transgenic mice, and distributed diffusely, mainly in the cytoplasm and minorly in the nucleus; (2) Under 40 times objective lenses, within 2 million pixel units, the average HBcAg-positive cell nucleus count within the liver was 92. 85 ± 50. 15, average area 70965.80 ± 37919. 20 pixels units , the average optical density of HBcAg-positive cell nucleus 0. 67 ±0. 03, the optical density ratio 2.37 ± 0. 09; (3) HBcAg was expressed unevenly, and higher expression was found around liver cell nucleus. Conclusion The four index items after statistical analysis show that there are no statistical difference in the average HBcAg-positive cell nucleus number, average positive-stained area, average optical density, and the optical density ratio between hepatic tissue portal area and central veins, and gender differences is not notable.
