亚硒酸钠对荷瘤小鼠体内硒含量,GSH-Px活力和MDA含量的影响

摘    要:目的:探讨荷瘤小鼠体内硒水平,谷胱甘肽过氧化物酶(GSH-Px)活力及丙二醛(MDA)含量的变化和亚硒酸钠对这种变化的影响。方法:采用DAN荧光法、DTNB比色法及TBA荧光法分别测定荷瘤及正常小鼠体内微量元素硒的含量、GSH-Px活力及过氧化脂质产物MDA的含量。结果:发现荷瘤小鼠体内全血及肝组织中硒的含量、肝匀浆中GSH-px活力均明显低于正常小鼠。亚硒酸钠虽然在该实验剂量下对S-180肉瘤的生长没有显著的抑制作用,但能纠正荷瘤小鼠全血及肝组织中硒含量的下降,增加全血及肝组织中GSH-Px活力,降低血浆中MDA的含量,对抗CCl4引起的肝组织中GSH-px活力下降。结论:亚硒酸钠能对抗肿瘤对机体抗氧化能力的削弱作用,这时肿瘤的预防和治疗有一定意义。

Abstract:Objective:To investigate the effects of sodium selenite on the level of selenium,activity of glutathione peroxidase(GSH-Px) and contents of lipid peroxide metabolites malondialdehyde(MDA) in mice with transplanted sarcomas. Methods : The level of selenium,activity of GSH-Px and contents of MDA in mice were determined by 2, 3-diaminonaphthalene(DAN) fluorophotometric,5,5'-dithionbis(2-nitrobenzoic acid)(DTNB) photometric and 2-thiobarbituric acid (TBA) fluorophotometric methods,respectively. Results: The level of selenium and activity of GSH-Px in blood and hepatic tissues of mice with transplanted sarcomas were significantly lower than those of normal mice. The oral administration of sodium selenite increased the level of selenium and activity of GSH-Px in blood and hepatic tissues markedly with a corresponding decrease in the contents of MDA in blood plasma of all mice. Sodium selenite could also retrieve the activity of GSH-Px in hepatic tissues effectively that had been inhibited by intraperitoneal administration of carbon tetrachloride (CCl4).In these lower experimental dosage, sodium selenite possessed no inhibitory effects on the growth of S-180 sarcomas. Conclusion : The results suggest that sodium selenite may increase the antioxidant activities impaired by transplanted sarcomas in mice and it may be useful in the prevention and treatment of carcinous diseases.
