丹曲林对糖尿病兔心房重构及氧化应激的影响

摘    要:目的本研究旨在探讨丹曲林对四氧嘧啶诱导的糖尿病家兔心房肌结构重构及体内氧化应激水平的影响。方法应用四氧嘧啶成功诱导糖尿病家兔24只,随机分为糖尿病组和丹曲林干预组,12只正常家兔为对照组,分别测定氧化应激指标一氧化氮(NO)、丙二醛(MDA)、谷胱甘肽(GSH)、超氧化物歧化酶(SOD)及胶原容积分数。结果与正常对照组相比,糖尿病组兔的MDA水平明显升高,SOD水平有降低趋势,丹曲林干预组SOD高于糖尿病组,而MDA水平明显低于糖尿病组,两组间有统计学差异(P〈0.05),HE及MASSON染色示糖尿病家兔左房肌细胞较正常组排列紊乱,炎症浸润,纤维化明显,而丹曲林干预组炎性改变及纤维化程度均减轻,两组比较有统计学差异(P〈0.05)。结论丹曲林干预组体内氧化应激水平及心房肌纤维化程度均较糖尿病组家兔减轻,提示丹曲林能够拮抗糖尿病家兔氧化应激反应及心房肌纤维化。

The Effect of Dantrolene on the Atrial Structural Remodeling and Oxidative Stress in Alloxan-induced Rabbits

Abstract:Objective To investigate the influence of dantrolene on the level of GSH,SOD,NO,MDA and collagen in left atrium in alloxan-induced rabbit model. Methods Twenty-four alloxan induced diabetic rabbits were divided into two groups (n=12): Diabetic group(D) and group diabetic rabbits receiving Dantrolene (DR). 12 normal rabbits were assigned as control group. All animals were mea- sured level of SOD, MDA, NO and GSH and left atrial collagen volume fraction. Results SOD level decreased and MDA level increased in diabetic rabbits group compared with control group(P〈0.05 ),while in dantrolene group the parameters didn't have significant difference compared with control group,but they had significant difference compared with diabetes group (P〈0.05).Masson stained of left atriai tissure showed that LACVF in diabetic rabbits was more higher than normal but in dantrolene group it was decreased (P〈0.05). Conclusion The study shows that dantrolene may reduces oxidative stress in the diabetic rabbits and inhibits fibrosis process in Left atrium of the diabetic rabbits, and dantrolene may be have potential effect on preventing atrial flatter process.
