Abstract:AIM To study the protective effect and mechanism of aqueous extract of Dalbergia odorifera(DOA)and volatile oil of Dalbergia odorifera(DOO)on isoprenaline?induced acute myocardial ischemia in rats.METHODS Myocardial ischemia rat model was induced by subcutaneous injections of isproterenol(ISO,30 mg/kg).Seventy rats were randomly divided into normal group,model group,propranolol positive control group(30 mg/kg),low dose DOA group(160 mg/kg),high dose DOA group(320 mg/kg),low dose DOO group(36 mg/kg),high dose DOO group(72 mg/kg),ten rats in each group were given intragastric administration once daily for five days.The rats had their changes in ST segment monitored by electrocardiogram,their serum myocardial enzymes detected by automatic biochemical analyzer,their degree of myocardial pathological damage observed by HE staining,their high?energy phosphate molecules in cardiac tissues detected by HPLC and their gene expression of AMPK,PKM2mRNA detected by PCR.RESULTS DOA and DOO significantly inhibited the ST segment changes caused by ISO(P<0.01),reduced the level of serum myocardial enzymes in myocardial ischemia rats(P<0.01);improved the myocardial tissue rupture of myocardial fibers and inflammatory cell infiltration;and increased the level of high?energy phosphate molecules in heart tissue.DOO enhanced AMPK,PKM2,CPT1 gene expression(P<0.01),while DOA inhibited PKM2,PFKFB3 gene expression(P<0.01).CONCLUSION Both being protective to acute myocardial ischemia in rats,DOA may activate the AMPK signaling pathway and enhance fatty acid oxidation to adjust myocardial energy metabolism,and DOO may inhibit glycolysis to adjust myocardial energy metabolism.
