| 摘 要： | 目的探讨葡萄糖调节蛋白78(GRP78)mRNA及蛋白在肝癌和癌旁肝硬化组织中的表达情况及其与肝癌发生的关系。方法肝癌手术切除标本30份,应用半定量逆转录—聚合酶链反应(RT-PCR)分别检测肝癌组织和癌旁肝硬化组织中GRP78mRNA的相对表达量,并通过免疫组化SP法检测肝癌组织和癌旁肝硬化组织中GRP78蛋白的表达情况。结果①半定量RT-PCR显示,GRP78mRNA在所有肝癌组织中均呈高表达,相对表达量为1.7037±0.8501,而在癌旁肝硬化组织中呈低表达或不表达,相对表达量为0.2742±0.3452,2组之间差异具有统计学意义(P0.01)。②免疫组织化学及图像分析结果显示,在肝癌组织和癌旁肝硬化组织中GRP78蛋白平均灰度扫描值分别为171.22±20.9705及139.53±20.2320,2组之间亦存在统计学意义(P0.01)。结论GRP78在肝癌中的表达明显增高,可能在肝癌的发生、发展过程中具有重要意义。
