mTOR、S6K1在双胍类降糖药物改善2型糖尿病中的作用研究

摘    要:目的 探讨mTOR、S6K1在双胍类降糖药物改善2型糖尿病中的作用和意义.方法 选取本院2015年3月至2017年3月收治的100例糖尿病患者,随机分为观察一组和观察二组,观察一组患者给予常规胰岛素治疗,观察二组患者在此基础上接受二甲双胍治疗3个月.选取同期健康体检者50例为对照组.治疗前后分别检测受试者血清中mTOR、S6K1信号通路的水平变化.结果 与对照组相比,观察一组和观察二组患者体内mTOR和S6K1水平显著升高,IL-6、TNF-α和IL-Iβ水平显著上升,差异具有统计学意义(P<0.05).观察一组和观察二组患者与治疗前相比,治疗后mTOR、S6K1、IL-6、TNF-α和IL-Iβ水平显著降低,差异具有统计学意义(P<0.05).与观察一组患者治疗后相比,观察二组患者二甲双胍治疗后,血清mTOR、S6K1、IL-6、TNF-α和IL-Iβ水平显著降低,差异具有统计学意义(P<0.05).患者血液mTOR和S6K1与其使用二甲双胍治疗的疗效存在显著的负相关关系,Person相关系数为负值,P<0.01.结论 双胍类降糖药物可以明显改善糖尿病患者的临床症状;mTOR、S6K1信号通路在糖尿病患者的疾病发展和临床治疗中发挥重要的作用.

Abstract:Objective To investigate the effect and significance of mTOR / S6K1 in the treatment of type 2 diabetes mellitus with biguanide and hypoglycemic agents.Methods From 2015 / 3 to 2017 / 3,100 patients with diabetes mellitus were randomly divided into two groups:observation group one and observation group two.The patients of group one were treated with the routine insulin therapy,while in addition,the group two received metformin treatment for 3 months.Fifty healthy subjects were also selected as the control group.The changes of mTOR / S6K1 signal pathway in serum were detected before and after treatments.Results Compared with the control group,the levels of mTOR and S6K1 were significantly increased and the levels of IL-6,TNF-o and IL-I3 were significantly increased as well in both observed groups (P < 0.05).The levels of mTOR,S6K1,IL-6,TNF-α and IL-I3 were significantly decreased after treatment for both groups(P < 0.05).The levels of serum mTOR,S6K1,IL-6,TNF-α and IL-Iβ were significantly decreased in the group two after treatment with metformin (P < 0.05).There was a significant negative correlation between the efficacy of mTOR and S6K1 in the treatment of patients treated with metformin,and the Person correlation coefficient was negative(P <0.01).Conclusion The double guanidine hypoglycemic agents can significantly improve the clinical symptoms of diabetic patients.The mTOR / S6K1 signaling pathway plays an important role in the development and clinical treatment of diabetic patients.
