摘    要:目的研究三氧化二砷(As2O3)对多发性骨髓瘤细胞株U266的作用。方法应用不同浓度的As2O3处理U266,采用细胞培养、流式细胞测定、原位杂交和酶联免疫吸附测定技术检测As2O3对U266的细胞活力、细胞凋亡、癌基因bcl-2表达和血管内皮细胞生长因子(VEGF)分泌的影响。结果As2O3可抑制U266的细胞活力,诱导细胞凋亡,抑制细胞bcl-2表达和VEGF的合成,VEGF可部分阻断As2O3的作用。结论As2O3可以通过多种机制发挥抗肿瘤作用。

Effect of arsenic trioxide on myeloma U266 cells

Abstract:Objective To investigate the effect of arsenic trioxide on multiple myeloma U266 cell line. Methods U266 cells were cultured with different concentrations of As2O3, and the effect of As2O3 on the vigor, apoptosis, expression of bcl-2, secretion of VEGF of U266 cells were detected by flow cytometry, situ hybridization, enzyme-linked immunosorbent assay. Results As2O3 could decrease the vigor, expression of bcl-2 and secretion of vascular endothelial cell growth factor (VEGF), and could induce apoptosis of U266 cells in dosage-dependent and time-dependent manners, and VEGF could partly block the effect of As2O3. Conclusion As2O3 can play antitumour role in multiple mechanisms.
