康莱特注射液诱发肾癌细胞凋亡及p53,bcl—2表达的研究

摘    要:目的;探讨康莱特注射液抗肿瘤的作用机制。方法:利用MTT法分析康莱特肾癌细胞的抑制作用,末端脱氧核苷酰转移酶法检测细胞凋亡,免疫组织化学法分析p53和bcl-2基因表达的影响。结果:康莱特抑制肾癌细胞的IC50为19.31μl/ml,5μl/ml,和10μl/ml康莱特注射液具有诱发细胞凋亡的作用,细胞凋亡分别为31.30%和89.76%,康莱特浓度继续增加时,细胞凋亡的数量反而减少,15μdispla

Abstract:Purpose: To study the apoptosis induced by Kang-Lai-Te injection (KLT)in renal cancer cell lines (GRC-1) Methods: The inhibitive effect of KLT on GRC-1 was measured by MTT Apoptosis was tested by TUNEL The expression of p53 and bcl-2 genes was determined by immunohistochemical staining Results: KLT could inhibit the GRC-1 cell growth, the IC50 value was 1931l/mlWhen it was exposed to 5l/ml and 10l/ml of KLT for 48 hours, the apoptotic percentages of renal cancer cell lines were 3130%and 8976%respectively, and it was less than 10%at the concentration of 1520l/ml The expression level of bcl-2 gene was lowered significantly at the concentration of 520l/ml than that of the control group Conclusion: KLT injection can induce apoptosis in renal cancer cells It may increase the p53 gene expression but inhibits the bcl-2 gene expression
