S100A4蛋白表达与胃癌侵袭转移关系的研究

目的：探讨Ｓ１００Ａ４蛋白产与胃癌侵袭转移的关系。方法：应用免疫组织化学方法检测６２例胃癌患者的胃癌组织和３８例癌旁正常粘膜、２３例淋巴结转移癌及胃癌纱ＭＫＮ４５、ＢＧＣ８２３中Ｓ１００Ａ４蛋白表达，Ｂｏｙｄｅｎ小室法检测胃癌细胞侵袭能力。结果：胃癌组织Ｓ１００Ａ４表达阳性率（７０．５９％）显著高于癌旁正常粘膜（１５．７９％）（Ｐ〈０．０１）。在弥温生长、低分化及未分化型、浸透浆膜、淋巴结转移阳性

Relationship between S100A4 Protein Expression and Invasion and Metastasis of Gastric Cancer

Objective: To study the relationship between the level of S100A4 expression and invasion and metastasis of gastric cancer. Methods: S100A4 protein expression was determined by immunohistochemical method in samples of cancerous tissue from 62 gastric cancer patients and 38 patient's corresponding non-cancerous mucosa, 23 metastatic lymph nodes, and the expressions of S100A4 in gastric cancer cell lines MKN45, BGC823. In vitro invasive capacity of gastric cancer cells were determined by Boyden chamber method. Results: Positive rate of S100A4 protein in gastric cancer (70.96%) was significantly higher than that of non-cancerous mucosa (15.79%)( P < 0 .01). The S100A4 expressions in diffusely growing pattern, poorly differentiated, undifferentiated, serosa invaded, positive lymph node metastasis and multiple lymph nodes metastases ( >5 nodes) gastric cancer, were significantly higher than that in other type gastric cancer ( P < 0.01 or P < 0.05). Positive rate of S100A4 in metastatic tumor of LN (100%) was markedly higher than that in corresponding primary tumor (15.79%)( P < 0.05). The invasive capacity of MKN45 cells was higher than that of BGC823 cells ( P < 0.01), and the S100A4 expression was also significantly higher than that in BGC823 cells. Conclusion: The increase of S100A4 expression in gastric cancer and gastric cancer cell lines are correlated with invasiveness and metastasis of gastric cancer.
