高效液相色谱法测定复方夏天无片中原阿片碱和苯甲酰乌头原碱含量

目的建立同时测定复方夏天无片中原阿片碱和苯甲酰乌头原碱含量的高效液相色谱(HPLC)法。方法色谱柱为Agilent Zorbax Eclipse SB-C18柱(250 mm×4.6 mm,3.5μm),流动相为1%冰乙酸溶液(A)-乙腈(B),梯度洗脱(0~5 min时94%A,5~27 min时94%A→58%A,27~30 min时58%A→94%A,30~35 min时94%A),柱温为30℃,测定波长为265 nm,流速为1.0 m L/min,进样量为25μL,外标法定量。结果原阿片碱和苯甲酰乌头原碱质量浓度在0.25~10.0μg/m L范围内与峰面积线性关系良好(r>0.995,n=6);平均加样回收率分别为97.47%和100.50%,RSD分别为4.37%和4.31%(n=6)。结论该方法操作简便、快速、准确、重复性好,可用于同时测定复方夏天无片中原阿片碱和苯甲酰乌头原碱的含量。

Content Determination of Protopine and Benzoyl Aconitine in Compound Xiatianwu Tablets by HPLC

Objective To establish a high performance liquid chromatography(HPLC)method for simultaneous determination contents of protopine and benzoylaconitine in Compound Xiatianwu Tablets.Methods Samples were seperated in Agilent Zorbax Eclipse SB-C18 column with column temperature 30℃and detected at 265 nm wave length.The flowing rate was 1.0 m L/min,with injection volume of25μL.Gradient elution program(0-5 min 94%A,5-27 min 94%A→58%A,27-30 min 58%A→94%A,30-35 min 94%A),external standard method was selected as quantitative.Results protopine and benzoylaconitine showed good linear relationships in the concentration range 0.25-10.0μg/m L(r>0.995,n=6).Average recoveries of protopine and benzoylaconitine were 97.47%and100.50%,repectively.RSDs of repeatability,precision and stability tests were 4.37%and 4.31%,respectively(n=6).Conclusion This method has the advantages of simple operation,rapid,accurate and good repeatability,which can be used for the simultaneous determination of contents of protopine and benzoylaconitine in Compound Xiatianwu Tablets.
