苯巴比妥对癫癎大鼠海马苔藓纤维突触重组和认知发育的影响

摘    要:目的探讨苯巴比妥(PB)对不同发育期癫痫大鼠海马内苔藓纤维(MF)发芽与认知功能的影响。方法Wistar大鼠按出生后日龄分组(P5、P15、P25、P60),以红藻氨酸(KA)制备癫痫模型,实验结束取脑制片,行Timm染色,观察海马齿状回内分子层染色改变。认知功能测验采用跳台法。结果KA致痫对P5、P15组海马内MF发芽现象、大鼠学习记忆功能无明显影响,但可使P25、P60组大鼠海马内MF发芽现象加剧,伴学习、记忆成绩明显下降,PB可加剧这一现象。PB对未致痫大鼠海马内MF发芽现象和学习记忆功能无明显影响。结论海马MF突触重组是PB影响不同发育时期癫痫大鼠学习记忆功能的重要原因。

Abstract:Objective To investigate the effects of phenobarbital(PB) on cognition and synaptic reorganization of hippocampal mossy fibers(MF) in developmental epileptic rats. Methods Wistar rats according to postnatal day P5, P15, P25, P60 divided into four groups(each group 40 rats). Epileptic models were established by injection of kainic acid(KA). Timm's silver sulfide staining were used to examine the MF sprouting in hippocampal inner molecular layer. Using step down test tested the cognitive function. Results KA administration show no significant different effects on P5 and P15, but elicit the increase of the MF sprouting and cognitive disturbances in older rat(P25 and P60) and PB injection exacerbated the disturbances. Conclusion The synaptic reorganization of hippocampal MF is an important factor in the cognitive disturbances. PB exacerbated the disturbances.
