摘    要:目的 探讨重组人促红细胞生成素(rHuEPO)对大鼠肝切除后肝再生的作用.方法 建立大鼠70%肝切除模型,术前30 min实验组经门静脉灌注rHuEPO 4000 IU/kg,对照组给予同等剂量的生理盐水.术后1 d,2 d、4 d,7 d两组各随机处死大鼠6只,采用全自动分析生化仪检测血清丙氨酸氨基转氨酶(ALT),免疫组化方法 检测肝组织增殖细胞核抗原(PCNA)、Ki-67表达,原位末端脱氧核苷酸转移酶法(TUNEL法)检测细胞凋亡.结果 肝切除术后1 d、2 d实验组ALT明显低于对照组(P<0.05),术后1 d、2 d,4 d实验组Ki-67标记指数明显高于对照组(P<0.05),术后1d、2 d,4 d,7 d实验组PCNA标记指数明显高于对照组(P<0.05),术后1d,2 d实验组凋亡细胞数明显低于对照组(P<0.05).结论 rHuEPO促进大鼠肝切除后肝再生.

Abstract:Objective To investigate the effects of recombinant human erythropoietin (rHuEPO) on the hepatic regeneration in rats after partial hepatectomy. Methods Rats undergoing 70% hepateetomy received an intraportalvenous administration ( i. p. ) of rHuEPO (4000 IU/kg) prior to resection, control group were treated with surgery and saline injection only. Six rats in each group were selected randomly and killed on the 1 st, 2nd ,4th and 7th day after surgery. The serum levels of alanine aminotransferase(ALT) were assessed ,the proliferating cell nuclear antigen ( PCNA ) , Ki-67 expression and hepatoeellular apoptosis were detected by immunohistochemistry and TUNEL assay. Results Compared with control group, the serum ALT of EPO group was significantly lower on the 1st and 2nd day after surgery (P 〈0. 05). Ki-67 labeling index was significantly higher in EPO group than that in control group on the lst,2nd and 4th day after surgery (P 〈0. 05). PCNA labeling index was significantly higher in EPO group than in control group on the 1st, 2nd,4th and 7th day after surgery( P 〈 0. 05 ). The level of apoptotic index was significantly lower in EPO group than in control group from day 1 to day 2 (P 〈 0. 05 ). Conclusions rHuEPO may promote the hepatic regeneration in rats after partial hepatectomy.
