电针对脑梗死大鼠海马齿状回Notch1调控作用的研究

摘    要:目的:观察电针对脑梗死大鼠海马齿状回Notch1的调控作用。方法:采用线栓法成功制备雄性SD大鼠右侧大脑中动脉闭塞(MCAO)模型大鼠,随机分为电针组、模型组和假手术组。在造模成功后第3天开始电针大鼠"百会"、"人中"穴15分钟/次,直至各个取材时间点。对各组在电针治疗各时间点行Morris水迷宫测试,采用免疫印迹法检测大鼠梗死侧海马齿状回区Notch1蛋白表达量。结果:与相同时间点模型组比较,电针组的Morris水迷宫测试差异有显著性意义(P0.05)。在梗死侧海马齿状回区域,电针组在治疗第3天和第7天时Notch1蛋白表达量较模型组相同时间点增高,差异具有显著性意义(P0.05),且第7天时达到高峰,在第14天时仍高于模型组,但差异无显著性意义(P0.05)。结论:电针能上调局灶性脑梗死大鼠梗死侧海马齿状回区Notch1蛋白表达量,改善脑梗死大鼠的认知功能,从而减轻脑梗死对神经元的损伤,达到治疗脑梗死功能障碍的疗效。

A study on the regulation effect of electroacupuncture on Notch1 at dentate gyrus in rats with acute cerebral infarction

Abstract:Abstract Objective: To investigate the regulation effect of electroacupuncture (EA) on Notch1 at dentate gyrus in rats with acute cerebral infarction. Method: Intraluminal middle cerebral artery occlusion (MCAO) was performed to establish the cerebral ischemia model. The male adult SD rats were randomly divided into the electroacupuncture group (EA group), the ischemia model group (MCAO group), and the sham operation group (sham group). EA was applied at Baihui and Renzhong after operation, 15min once a day from operation to the day sacrificed. Each group rats were assessed the Morris water maze test before and after intervention. The expression of Notch1 protein in hippocampus was detected by Western Blotting. Result: Compared with the model group, EA at Baihui and Renzhong acupoints alleviated cognitive impairment significantly and the expression of Notch1 protein in hippocampal of EA group was significantly increased. Conclusion: EA at Baihui and Renzhong acupoints can improve cognition and upregulate the expression of Notch1 of protein of hippocampal, so as to reduce the damage of cerebral ischemia on neurons, and provide the therapeutic effect on cerebral ischemia.
