应用分子杂交技术检验白细胞内HBV DNA

摘    要:本研究应用斑点分子杂交技术检测了157例慢性HBV感染者及80例一般人群(含20例献血员)的血清和白细胞内HBV DNA的状况。在157例慢性HBV感染者中血清HBV DNA阳性率为39%,白细胞内HBV DNA为36%,两者同时阳性者为18%。而在60例血清HBV DNA阳性者中白细胞内HBV DNA同时阳性者为48%,血清HBV DNA阴性的93例中仍有30%的病例白细胞内HBV DNA为阳性。说明在血清HBV DNA阴性的慢性HBV感染者中仍有1/3的病例的白细胞内存在HBVDNA。80例一般人群经RIA法筛选发现:单纯HBsAg阳性6例,其中3例白细胞内HBV DNA阳性。HBsAg与HBeAg同时阳性者4例,其白细胞内HBV DNA均为阳性。单纯抗-HBs阳性的24例中仅1例白细胞内存在HBV DNA。单纯抗-HBc阳性者为12例,白细胞内HBV DNA无1例阳性。值得注意的是,在血清HBV标志全阴性的34例中也发现了2例白细胞内HBV DNA阳性。由于HBV可以感染人的白细胞,而且影响白细胞的免疫功能,其在形成HBV感染的慢性化上可能起着重要的作用,因而在肝炎的转归、流行病学的意义及发病机制等问题上它的作用尚需进一步探讨。

Abstract:The HBV DNA status in sera and leucocyte of 157 patients intected with chronic HBV and 80 general population (include 20 blood donors) were studied. Of the 157 cases, 39% were serum HBV DNA positive, 36% were leucocyte HBV DNA Positive, and 18% serum HBV DNA and leucocyte HBV DNA positive, Of the 60 patients with serum HBV DNA positive, 48% wereleucocyte HBV DNA positive. Still, 30% were leucocyte HBV DNA positiveout of the 93 patients with serum HBV DNA negative, which impliesthat 1/3 chronic-HBV-infected patients with serum HBV DNA negativewere leucocyte HBV DNA positive, By screening the 80 general populationwith RIA method, it was found that 3 of the 6 patients with HBsAgpositive were leucocyte HBV DNA positive, 4 cases with both HBsAg andHBeAg positive were leucocyte HBV DNA positive. Of the 24 cases withanti-HBs positive, only 1 was leucocyte HBV DNA positive. It should beespecially noted that out of the 34 samples with all the serological markersnegative, 2 were leucocyte HBV DNA positive. Sice HBV not only infects leucocyte but also interfere with the immunological functions of the white blood cells, it pernaps plays an important role in the formation of HBV chronic infection. So, its functions in the hepatitisrecovery, epidemiological importance and disease mechanism should be furtherstudied.
