一氧化氮与肝硬化心肌病的关系研究

摘    要:为探讨肝硬化心肌病的发生机制,应用放射性免疫分析法(RIA)测定胆管结扎(BDL)诱导的胆汁性肝硬化大鼠和假手术大鼠血浆及心脏组织匀浆中一氧化氮(NO)依赖性环-磷酸鸟苷(cGMP)深度,采用免疫组织化学法对上述2组大鼠心脏内皮型一氧化氮合酶(eNOS)和诱导型一氧化氮合酶(iNOS)的表达进行比较性研究。结果显示:肝硬化大鼠血浆和心脏组织匀浆中cGMP水平显著高于假手术组(P<0.01);肝硬化和假手术大鼠心脏eNOS均呈阳性表达,2组之间无显著性差异,而iNOS在肝硬化大鼠心脏呈强阳性表达,在假手术组则为阴性。提示肝硬化大鼠血浆和心脏组织中NO含量显著增加,而增加的NO主要由iNOS诱导产生,并由此推断一氧化氮途径可能在肝硬化心肌病的发生中起重要作用。

Abstract:To investigate the pathogenesis of cirrhotic cardiomyopathy, concentration of cGMP in plasma and homogenates of cardiac tissue of cirrhotic rats (CIR) induced by bile duct ligation (BDL) and sham-operated rats (SO) was measured by radioimmunoassay (RIA). Controlled studies on the expression of eNOS and iNOS in heart of two groups of rats were undertaken using immunohistochemistry. It was found that the level of cGMP in plasma and homogenates of cardiac tissue of CIR rats was significantly higher than in the SO rats (P<0.01); The expression of eNOS in heart of two groups of rats was all positive, the expression of iNOS in heart of CIR rats was strong positive, but it was negative in SO rats. It was suggested that content of NO in plasma and cardiac tissue of CIR rats was significantly increased, and the increased NO was induced by iNOS. It was concluded that NO-approach may play an important role in the pathogenesis of cirrhotic cardiomyopathy.
