甲基硝基亚硝基胍对日本血吸虫成虫培养细胞骨架作用的研究

目的 通过观察N－甲基－N－硝基－N－亚硝基胍（MNNG）诱导日本血吸虫培养细胞后骨架的改变，研究MNNG对培养细胞转化、增殖的作用。方法 将日本血吸虫成虫培养细胞接种于小盖玻片上，培养1周时用浓度为3mg/L的MNNG处理48h，然后用RPMI－1640含20％小牛血清附加常量抗菌素的培养基培养2周，再换用RPMI－1640含5％小牛血清的培养基继续培养，用考马斯亮兰染色法和鬼笔环肽荧光染色法显示培养细胞骨架。结果 MNNG诱导后，在培养第4、5周时部分培养细胞骨架排列紊乱，微丝集聚于细胞边缘。结论 MNNG有一定诱导日本血吸虫成虫培养细胞转化、促进其分裂的作用。

Studies on the effect of MNNG on cytoskeleton of cultured cells from adult Schistosoma japonicum

Objective To study the effect of MNNG on reproduction and transformation of cultured cells from adult Schistosoma japonicum by observing changes of cytoskeleton of the cells.Methods Shisotomal cells were inoculated on small glass coverslips and were cultured for one week.Then the cells were treated with N Methy N Nitro N Nirtoso guanidine(MNNG) with concentration of 3 mg/L for 48 h.Afterwards,the cells were cultured with RPMI 1640 containing 20% of calf serum and antibiotics at normal dose for two weeks and then with RPMI 1640 containing 5% of calf serum.The cells were observed with Comassie blue and Rhodamin Phalloidine Microfilaments staining.Results In some cells treated by MNNG the arrangement of cytoskeleton appeared disorder and microfilament assembled together in cellular edge in the fourth and fifth weeks.Conclusions MNNG had effect to some extent on inducing the transformation of the cells from adult Schistosoma japonicum.
