氯化锂-匹罗卡品致痫幼鼠成年后少突胶质细胞转录因子Olig2在脑白质区的表达

摘    要:目的探讨氯化锂-匹罗卡品(lithium-pilocarpine,LI-PILO)点燃幼鼠癫痫持续状态(status epilepticus,SE),成年后发展为自发反复性癫痫发作(spontaneous recurrent epileptic seizure,SRS)的大鼠脑内转录因子Olig2的表达。方法采用氯化锂-匹罗卡品腹腔注射诱导幼鼠SE模型,应用多道生理信号采集处理系统检测大鼠脑电图(electroencephalogram,EEG)痫样活动波和Nissl染色方法观察神经元鉴定,证实SRS模型大鼠诱导成功;应用免疫组织化学染色技术观察少突胶质细胞(oligodendrocyte,OL)转录因子Olig2变化。结果 SRS模型组脑电图与正常对照组相比,可见频发尖波、棘-慢、尖-慢综合波;海马CA1、CA3和齿状回区Nissl染色显示神经元数目减少,部分细胞变性和坏死。SRS模型组在胼胝体与扣带回区域内Olig2的免疫组织化学染色细胞数量明显减少,与正常对照组相比有显著差异。结论氯化锂-匹罗卡品致痫幼鼠,成年后仍伴有慢性自发反复性癫痫发作的脑内少突胶质细胞转录因子Olig2的表达明显降低。

Abstract:Objective To investigate the expression of transcription factor Olig2 in the brain of mature rats with spontaneous recurrent epileptic seizure (SRS) after lithium-pilocarpine-induced status epilepticus (SE) in their infancy.Methods SE model was induced by lithium-pilocarpine in young rats.Multiple Channel Physiological Signal Collecting and Processing System was used to monitor the abnormal wave of epilepsy on the electroencephalogram (EEG).Nissl staining was used to observe the alteration of the neuron in the hippocampus.The expressions of transcription factor Olig2 were identified by histopathological and immunohistochemical methods.Results EEG and Nissl staining confirmed the successful induction of SRS model by comparing the wave and cell morphology of control and SRS groups.The epileptic waves,frequent sharp waves,spike-and-slow wave and sharp-and-slow wave complexes were found in SRS model group.The number of neurons was decreased and partial cells showed denaturation or necrosis in the areas of CA1,CA3 and white matter in the hippocampus.Immunohistochemical staining showed Olig2-positive cells in SRS model group were dramatically decreased in the callosum corpus (cc) and cingulate gyrus (cg) areas.Conclusion Expression of Olig2 in oligodendrocytes decreases significantly in the rats that continued to have spontaneous recurrent seizures into adulthood after lithium-pilocarpine-induced seizure in their infancy.
