摘    要:目的 明确γ射线照射致淋巴细胞DNA损伤的量效关系,拟合曲线方程。方法 采用聚苯乙烯凹槽单层铺胶后进行碱性单细胞凝胶电泳,观察γ射线外照射引起的人外周血淋巴细胞DNA损伤效应。应用IPP软件采集“彗星”尾长,利用SPSS软件进行数据分析和曲线拟合。结果 γ射线照射导致淋巴细胞DNA单链断裂,DNA断片电泳图像清晰。“彗星”尾长与照射剂量正相关(r=0.907,P<0.01),量效曲线的拟合方程为y=109.369319-40.258796x 13.636240x~2(P<0.001)。结论 一元二次方程可以很好反映淋巴细胞DNA断片迁移长度与γ射线照射剂量之间的量效曲线。

Abstract:Purpose To investigate the relationship of the radiation dose to DNA damage in lymphocytes and its curve estimation. Methods The radiation-induced DNA damage in human peripheral blood lymphocytes was e-valuated using alkaline single cell gel electrophoresis by filled the polystyrene ponds with mono-layer gel embedded lymphocytes. The length of "comet tail" was measured with IPP software and the data were analyzed by SPSS. Results The DNA single strand breaks in lymphocytes were caused by irradiation and its electrophoresis images were dear. The tail length was positive correlation( r = 0.907, P< 0.01) with dose and its curve-fit equation was y = 109.369319 - 40.258796x 13.636240x2(P<0.001). Conclusions The quadratic model is fit for the curvilinear relationship of the migration length to radiation dose.
