| 摘 要： | 目的 :研究氧化低密度脂蛋白 (0x -LDL)引起血管平滑肌细胞 (VSMCs)凋亡的效应及其凋亡细胞周期分析。方法 :采用电镜、流式细胞仪等观察Ox -LDL诱导VSMCs凋亡的细胞形态学改变及细胞周期变化。结果 :VSMCs在Ox -LDL(2 0mg/L)作用下 ,电镜下见凋亡细胞呈现核固缩凋亡小体的形态学特征。流式细胞仪检测发现Ox -LDL(2 0mg/L)引起细胞凋亡指数明显高于N -LDL(10倍 )。细胞增殖周期分析可见 ,Ox -LDL处理VSMCs 2 4h ,随着浓度的增加 ,G1期细胞百分率逐渐下降 ,而G2 /M期细胞百分率逐渐升高 ,提示Ox-LDL能将细胞阻滞在G2 /M期。结论 OX -LDL能诱导VSMCs凋亡 ,VSMCs的凋亡与细胞分裂期阻滞密切相关。

| Abstract: | Objective:To investigate the apoptosis of vascular smooth muscle cell of rats induced by oxidized low density lipoprotein (Ox-LDL) and the mechanism of its action. Method:The cell morphology, flow cytometry were performed for 0x-LDL-inducing apoptosis of VSMCs in rats. Results:The VSMCs treated with Ox-LDL showed cell shrikage, condensation or fragmentation of chromosomes and apoptotic body. Flow cytometry detection of DNA by using propidium iodide (PI) labelling showed G 1 phase cells decreased, sub-G 1phase cells (apoptotic cells) appeared. Growth cycle of apoptotic cell appeared that percentage rate of G 1 phase cell decreased gradually, but G 2/M phase cell increased as Ox-LDL increased. Conclusion: Ox-LDL induced apoptosis of VSMCs, The induction of apoptosis of aortic VSMCs in mice by 0x-LDL was closely associated with mitotic arrest of cell cycle. |
