唑来膦酸对胃癌细胞株SGC-7901体外抗增殖及对VEGF的影响

目的研究唑来膦酸对胃癌细胞株SGC-7901的体外增殖抑制作用及对血管内皮生长因子（VEGF）的影响。方法采用CCK-8试验检测唑来膦酸对胃癌细胞株SGC-7901的生长抑制作用;酶联免疫吸附试验（ELISA）和半定量RT-PCR法检测药物对VEGF表达水平的变化。结果 0.2～3.2μg/L的唑来膦酸能显著抑制SGC-7901细胞的增殖,且具有时间及剂量依赖性,不同药物浓度作用后,胃癌细胞株VEGF的蛋白及基因表达水平均明显降低（P〈005）,具有浓度依赖性。结论唑来膦酸能抑制胃癌细胞株SGC-7901的生长增殖,其作用机制可能为下调VEGF表达。

Proliferation inhibition function of zoledronic acid on human gastric cancer cell SGC-7901 and its influence on VEGF

Objective To investigate the proliferation inhibition function of zoledronic acid on human gastric cancer cell SGC-7901 and its influence on vascular endothelial growth factor（VEGF）. Methods CCK-8 cell viability assay was performed to investigate the effect of zoledronic acid on cell proliferation. The expression of VEGF was examined by ELISA and semi-quantitative RT-PCR. Results Zoledronic acid inhibited the proliferation of SGC-7901 cells at a dose-and-time dependent manner（concentration range： 0.2～3.2 μ g/L）. Meanwhile, ELISA and semi-quantitative RT-PCR results showed that the expression of VEGF was down-regulated at a dose-dependent fashion in SGC-7901 cells. Conclusion Zoledronic acid might inhibit the proliferation of SGC-7901 cells through down-regulation of VEGF.
