葛根素对左室局部AngⅡ水平的影响及对左室肥厚的逆转作用

摘    要:目的探讨葛根素逆转肾性高血压大鼠左室肥厚是否与调节血浆及左室局部生物活性因子AngⅡ、ET-1和NO有关。方法应用两肾一夹塑造肾性高血压大鼠模型。葛根素以200、100及50mg/kg剂量分别腹腔注射给药6周。放免法测ET-1和AngⅡ浓度,硝酸还原酶法测定NO含量,考马斯亮蓝法进行组织蛋白定量。称量大鼠体质量和左室湿重,计算左室质量指数(LVMI)。结果葛根素各剂量均可影响LVMI,高、中剂量组血浆及左室局部AngⅡ和ET-1明显降低,NO明显升高,与模型组相比均有显著意义(P<0.01或P<0.05)。低剂量组左室局部AngⅡ浓度降低,左室局部其他指标没有明显改变。结论葛根素保护心肌与调节左室局部生物活性因子含量有关。

Abstract:Objective To explore whether puerarin reversing left ventricular hypertrophy is related to changes on the levels of AngⅡ,ET-1 and NO in plasma and left cardiac ventricle.Methods A rat model of renal hypertension was induced by two-kidney,one-clip.Puerarin was intraperitoneally administered at a concentration of 200,100 and 50 mg/kg(n=8 each)for six weeks respectively.ET-1 and AngⅡ were measured by radioimmunoassay.NO was measured by nitrate reductase.Protein content of the tissue was determined by a Coomassie blue dye-binding assay.Left ventricular mass index (LVMI) calculated by dividing left ventricular mass in milligrams by rat weight in grams.Results Puerarin with three different doses significantly decreased LVMI compared with model group(P<0.01 or P<0.05).High-and middle-dose puerarin significantly reduced contents of AngⅡ and ET-1(P<0.01 or P<0.05),but increased that of NO markedly in plasma and left cardiac ventricle (P<0.01 or P<0.05).Low dose puerarin decreased only the level of Ang Ⅱ in cardiac tissue without obviously changing other biologically active factors mentioned above.Conclusion Puerarin′s protective effect on myocardium is related to its regulatory role for the levels of local myocardial AngⅡ,ET-1 and NO.
