摘    要:目的建立测定联磺甲氧苄啶片含量的高效液相色谱(HPLC)法。方法采用Zorbax C18柱(150mm×4.6mm,5μm)为色谱柱,以水-乙腈-三乙胺(799:200:1,用氢氧化钠试液或冰醋酸调节pH值至5.9)为流动相,检测波长为240nm,流速为1.0mL/min,进样量为20μL,枉温为30℃。结果磺胺嘧啶和磺胺甲嗯唑质量浓度在5~100μg/mL范围内,甲氧苄啶质量浓度在2~40μg/mL范围内与峰面积线性关系良好;平均回收率磺胺嘧啶为99.91%(RSD为0.2%),磺胺甲嗯唑为99.76%(RSD为0.2%),甲氧苄啶为99.80%(RSD为0.4%)。结论HPLC法分离度好,快速、简便,可同时测定3种组分,可作为该产品的质量控制方法。

Abstract:Objective To establish the HPLC method for the determination of sulfadiazine in Sulfamethoxazole, Sulfadiazine and Trimethoprim Tablets. Methods The column packed with C18 Zorbax (150 mm ×4.6 mm, 5μm) was used. The mobile phase was water-acetonitrile- triethylamine (799 : 200 : 1, adjusting pH to 5.9 by sodium hydroxide reagent or glacial acetic acid) and the detection wavelength was 240 nm,the flow rate was 1.0 mL/min, the injection volume was 20 μL, the column temperature was 30℃. Results The calibration curve of sulfamethoxazole and sulfadiazine was linear in the range of 5 - 100 μg/mL,and trimethoprim was linear in the range of 2 -40 μg/mL. The average recovery rate was 99. 76% for sulfamethoxazole ( RSD =0. 2% ) ,99.91% for sulfadiazine ( RSD =0. 2% ) ,99.80% for trimethoprim (RSD =0. 4% ). Conclusion The method is simple, accurate, and suitable for the determination of the three constituents in the compound preparation.
