摘    要:目的 评价大鼠肝部分切除术后认知功能及β淀粉样蛋白(Ap)的变化.方法 健康雄性sD大鼠70只,体重200~250 g,随机分为3组:对照组(C组,n=12)、麻醉组(A组,n=29)和肝部分切除术组(PH组,n=29).A组和PH组于术后1 d(T1)和7 d(T3)时各取7只大鼠,采用Y型迷宫检测术后第1、2、3、7、8、9天时学习记忆能力;于术后1 d(T1)、3 d(T2)和7 d(T3)时取5只大鼠,取左侧海马,并断尾取血,测定Aβ表达;取右侧海马采用荧光定量PCR测定App mRNA表达.结果 与C组相比,PH组术后学习记忆能力下降,T1,3时血清Aβ浓度降低,T1-3,时海马Aβ含量升高,TI时App770mRNA含量升高(P<0.05);与A组相比,PH组术后学习记忆能力下降,T1,3时血清Aβ浓度降低,T1-3时海马Aβ含量升高,T1时App770 mRNA含量升高(P<0.05).结论 肝部分切除术后大鼠发生短期认知功能减退,可能与海马Aβ含量升高有关.

The changes in cognitive function and beta amyloid protein after partial hepatectomy in rats
Abstract:Objective To investigate the changes in cognitive function and the expression of beta amyloid protein(Aβ)and beta amyloid precursor protein(App)mRNA in the hippocampus and Aβ in serum after partial hepatectomy in rats.Methods Seventy healthy male SD rats weighing 200-250 g were randomly divided into 3 groups:group Ⅰ control(C,n=12);group Ⅱanesthesia(A,n=29)and group Ⅲ partial hepatectomy(PH,n=29).Seven animals in each group underwent Y-maze teat once a day for 3 consecutive days on 1st,2nd and 3rd day after anesthesia or operation and another 7 animals on 7th,8th and 9th day after anesthesia or operation.Five animals were killed on 1st day(T1),3rd day(T2)and 7th day(T3)after anesthesia or operation respectively.Bilateral hippocalnpi were removed for determination of content of Aβ and App mRNA and blood samples were taken for determination of Aβ concentration.Results (1)The number of error in the test was significantly increased and the number of active avoidance was decreased on the 2nd,3rd and 8th day after operation in PH group as compared with control(C)and anesthesia(A)group.(2)The content of Aβ in hippocampus was increased at Ti,T2 and T1 and the serum Aβ concentration was decreased at T1 and T3 in group PH as compared with group C and A.The content of App mRNA in hippocampus was decreased at T1 in group PH as compared with group C and A.Conclusion Partial hepatectomy can impair cognitive function after operation for a short time.The increase in the content of Aβ in hippocampus may play a role in the mechanism.
