海人藻酸致痫大鼠心肌细胞ERK表达的实验研究

摘    要:目的研究海人藻酸致痫大鼠心肌细胞ERK表达情况。方法采用立体定位仪下杏仁核内注射海人藻酸方法制备癫痫动物模型,在大鼠癫痫发作后不同时程,快速制备心脏组织标本,进行免疫组化及染色,用Image-ProPlus图像分析软件,计算大鼠心肌细胞ERK表达灰度值的均数和标准差,观察不同时程心肌细胞ERK表达情况。结果致癫痫组大鼠心肌细胞ERK表达于癫痫发作后1/2小时开始增加,灰度值为4735.86±172.43,2小时心肌组织ERK的表达达到高峰,灰度值为12764.71±178.45,与对照组相比P值均<0.05,随后逐渐下降;至癫痫发作6小时,心肌组织ERK表达基本消失,灰度值为2389.78±185.64,与对照组相比P>0.05。结论ERK在癫痫所引起的脑心综合征中有表达。

Abstract:Objective To study the ERK expression of myocardial cell in epilepsy rat induced by kainic acid. Methods Rat epilepsy model was prepared by injection of kainic acid into amygdala under three-dimensional positioning devices. At different time points after epilepsy spasm, heart tissue specimens were immediately prepared for immunohistochemistry and staining. Using Image-Pro Plus image analysis software, rat myocytes ERK expression gray value (mean + SD) was calculated. Results Myocardial cells ERK expression (gray value) in epilepsy rat began to increase since 0.5 h after epilepsy spasm (4735.86 ± 172.43). It reached peak at 2 hours after epilepsy spasm (12764.71 ± 178.45). Myocardial tissue ERK expression in epilepsy rat was significantly higher than in control rat (P 〈 0.05), then it decreased gradually; at 6h after spasm, the myocardial tissue ERK expression was as high as before spasm (P 〉 0.05). Conclusion ERK expression was increased in the brain-heart syndrome caused by epilepsy.
