罗红霉素对小鼠腹腔巨噬细胞产生IL-8的影响

摘    要:目的:研究罗红霉素(Roxithromycin,Rox)对小鼠腹腔巨噬细胞产生IL-8的影响。方法:采取体内BCG刺激体外LPS诱生IL-8,按常规方法收集纯化8-12周龄昆明种雄性小鼠腹腔巨噬细胞,用RPMl1640培养基培养,按所用药物Rox浓度和地塞米松(Dex)进行分组并设计对照组。用RIA试剂盒测定IL-8含量。结果:Rox三个剂量组均能显著抑制小鼠巨噬细胞分泌IL-8,且高剂量组抑制更明显,高剂量组Rox的作用强度与Dex相似。结论:Rox抑制IL-8等炎性因子的产生可能是其抗炎、免疫调节的主要机制之一。

Abstract:Aims: To study the effect of roxithromycin(Rox) on IL 8 of macrophage in abdominal cavity of mice.Methods:LPS induce IL 8 by BCG in vivo, collecting and purifing the macrophage in abdominal cavity of male mice which is 8-12 week's old with common methods, then culture the macrophage with RPMI1640, including the control and all kinds of concentration on Rox and Dex(dexamethasone) groups. Assay the IL-8 with RIA kits.Results:All three Rox groups can remarkably inhibit IL 8 which is secrected by the macrophage of mice and the high Rox group is more obviously, the action strength of the high Rox group is similar to the Dex group.Conclusions: One of the chief mechanism of antiinflamation and immunoregulation produced by Rox maybe is to inhibit IL 8 inflammatory factor etc.
