尿毒清颗粒对糖尿病大鼠肾脏炎症损伤的影响

摘    要:目的探讨尿毒清颗粒对糖尿病大鼠肾脏炎症损伤的保护作用。方法选用链脲佐菌素(STZ)诱导的糖尿病大鼠模型。实验分3组:正常对照组(N组)、糖尿病未干预组(D组)、尿毒清颗粒治疗组(Q组,2.6g·kg-1·d-1灌胃),于第4、8周末检测血肌酐(SCr)、尿肌酐(Ucr),计算肌酐清除率(Ccr),检测血清超敏C反应蛋白(hs—CRP)、肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6),光镜观察肾组织病理改变,免疫组织化学检测肾组织核因子xB(NF-kB)、单核细胞趋化蛋白1(MCP-1)表达,检测肾皮质MCP-1mRNA表达。结果尿毒清颗粒可以提高糖尿病大鼠Ccr(P〈0.01),改善肾脏病理损伤,减少血清hs-CRP、肾组织NF-kB、MCP-1表达(P〈0.1)5,P〈0.01),对血清TNF-α、IL-6影响不显著。结论炎症反应参与糖尿病大鼠肾脏损伤,尿毒清颗粒能降低血清及肾脏炎症介质表达,对糖尿病大鼠肾脏炎症损伤起保护作用。

Effects of Niaoduqing particles on inflammation injury in diabetic rats

Abstract:Objective To evaluate the preventive effects of Niaoduqing particles on inflammation injury in diabetic rats. Methods SD rats were divided into three groups: control group, diabetic group without intervention, Niaoduqing group (intragastric administration with Niaoduqing, 2. 6 g. kg-1. d-1 ). After 4 and 8 weeks, serum creatinine, urine creatinine and serum hs-CRP, TNF-alpha, IL-6 were measured. Renal patho- logic changes were observed under a light microscope. The expression of NF-kB and MCP-1 was detected by using immunohistochemistry,and the mRNA expression of MCP-1 in the renal cortex by real time-PCR. Re- suits STZ induced diabetes complicated by renal dysfunction with decreased level of creatinine clearance rate. Treatment with Niaoduqing particles improved renal function,ameliorated renal pathologic chan4ges,less- ened the level of serum hs-CRP and the expression of renal NF-kB, MCP-1 ,but the levels of serum TNF-al- pha and IL-6 were not influenced. Conclusions Inflammatory reaction was correlated with renal injury in diabetic rats. Niaoduqing treatment could reduce the expression of inflammation cytokines, and partly prevent iinflammation injury in diabetic rats.
