高效液相色谱法测定蒲公英颗粒中咖啡酸含量

摘    要:目的用高效液相色谱法测定蒲公英颗粒中咖啡酸的含量。方法采用C18柱,以磷酸盐缓冲液(取磷酸二氢钠1.56g,加水使溶解成1000mL,再加1%磷酸溶液调节pH值至3.8-4.0,即得)-甲醇(75:25)为流动相,检测波长为323nm。柱温为40℃,流速为1.0mL/min。结果咖啡酸质量浓度在19.54-58.62μg/mL范围内与峰面积具有良好的线性关系(r=0.9998),平均回收率为100.75%。RSD为1.0%。结论谊法操作简单,结果准确,可有效地控制蒲公英颗粒的质量。

Abstract:Objective To determine caffeic acid in Pugongying granules sodium dihydrogen phosphate dissolved in water to 1 000 mL, adjusting by HPLC. Methods Using C18 column and phosphonate buffer(l. 56 g with 1% phosphoric acid solution to pH was 3.8-4,0)- methanol (75:25) as mobile phase, the detection wavelength was 323 nm and the column temperature was 40℃. The flow rate was 1.0 mL/min. Results The calibration curve were linear in the range of 19. 54-58.62 μg/mL for caffeic acid (r=0.999 8).The average recovery rate was 100. 75%, RSD was 1.0%. Conclusion This method is simple and convenient for the operation. It is very accurate and very suitable for the quality control of Pugongying granules.
