HPLC同时测定氢定乳膏中氢化可的松和醋酸氯己定的含量

 目的建立测定氢定乳膏中氢化可的松和醋酸氯己定含量的方法。方法采用高效液相色谱法。分析色谱柱为Extend C₁₈(4.6 mm×150 mm,5μm),流动相为甲醇-水相(醋酸铵26 g和三乙胺37 mL,加水至1 000 mL,冰醋酸调pH至4.0)(55∶45)为流动相;流速为1.0 mL·min^-1;检测波长为254 nm;柱温为30℃;进样量为10μL。结果该方法不受处方中基质干扰,氢化可的松在0.1～0.6 mg·L^-1内和醋酸氯己定在0.02～0.12 mg·L^-1内,峰面积与质量浓度呈良好的线性关系(r_{hydrocortisone}=0.999 8,r_{chlohexidine acetate}=0.999 8),平均回收率:氢化可的松为99.6%、醋酸氯己定为99.2%。RSD分别为0.34%,1.41%(n=9)。结论该方法可用于氢定乳膏中氢化可的松和醋酸氯己定的含量测定。

Simultaneous Determination of Hydrocortisone and Chlohexidine Aacetate in Qingding Cream by HPLC

OBJECTIVE To develop a method for the determination of hydrocortisone and chlohexidine acetate in Qingding cream.METHODSWTBZ] A high-performance liquid chromatography method was used with Extend C₁₈ column.The mobile phase consisted of methanol-aqueous phase(ammonium acetate 26 g and triethylamine 37 mL,imbitition to 1 000 mL,adjust pH to 4.0 by glacial acetic acid)(55∶45) and the flow rate was 1.0 mL·min^-1,with the detection wavelength at 254 nm.RESULTS The matrix component in cream didn't interfere with determination of hydrocortisone and chlohexidine acetate.The calibration curve was linear within the range of 0.1～ 0.6 mg·L^-1 for hydrocortisone(r=0.999 8) and the range of 0.02 ～ 0.12 mg·L^-1 for chlohexidine acetate(r=0.999 8).The average recoveries of hydrocortisone and chlohexidine acetate were 99.6% and 99.2% respectively.RSD were 0.34% and 1.41%(n=9),respectively.CONCLUSION The method can be used for the determination of hydrocortisone and chlohexidine acetate in Qingding Cream.
