生物酶制剂JA对小鼠乳腺癌(MA782肿瘤)的消融治疗作用

摘    要:目的探讨生物酶制剂JA治疗小鼠乳腺癌的可行性和安全性,为临床应用提供依据。方法噻唑蓝(MTT)法检测不同浓度JA制剂对MA782肿瘤细胞的作用,电镜观察JA制剂作用后肿瘤细胞的超微结构;用MA782细胞建立小鼠乳腺癌动物模型,共60只,随机分成3组(G1~G3),分别行瘤体内注射生理盐水、无水乙醇、制剂JA各0.2ml,观察用药后肿瘤大小变化,于用药第1、第10天每组各处死8只小鼠,采血,检查肝肾功能,取肿块病检。结果1/40倍浓度以上JA制剂作用30min即可将90%以上肿瘤细胞杀死;肿瘤消融效果G3〉G2〉G1,G2组出现ALT、AST持续升高,G3组ALT、AST仅表现为一过性升高,其他各组间各指标差异无统计学意义。结论制剂JA对实验性小鼠乳腺癌(MA782肿瘤)有独特的消融效果,明显优于无水乙醇,是一种安全有效的生物酶消融制剂,具有潜在的临床应用前景。

Abstract:Objective To investigate the safety and feasibility of enzyme JA in the treatment of rat breast cancer. Methods Cytotoxinic effects of the enzyme JA at different concentrations on MA782 tumor cells were tested by MTT assay. Under electron microscopy, the ultrastructures of tumors treated with JA were observed. Rat breast cancer models in 60 animals were established with MA782 cells and randomly divided into 3 groups (G1-G3), being subjected to saline, absolute alcohol and JA 0.2 ml each. The size of tumor was observed after treatment. On the first and 10th day after treatment, 8 rats were killed in each group. The pathologic examination was performed and serum ALT/AST/Cr/BUN tested. Results More than 90 % tumor cells could be killed by 1/40 JA in 30 min. The ablation effect of G3 was better than G2. In G2 group, ALT and AST were continuously elevated, and in G3 group, ALT and AST were temporarily increased. Conclusion The ablation effect of intra-tumor injection of the enzyme JA into implanted rat breast cancer is evident, and it is more effective and safer than ethanol.
