广枣及其提取组分对神经细胞的保护作用

摘    要:目的研究广枣及其提取组分对拟衰老神经细胞的保护作用并初步探讨其机制。方法通过建立拟衰老神经元模型,采用水提醇沉的方法将广枣水提液再分离,得到沉淀大分子组分和上清小分子组分,并观察它们对拟衰老神经元的保护作用和其通过星型胶质细胞对拟衰老神经元的间接保护作用。结果广枣水提液、小分子组分和其作用的星型胶质细胞培养液均能显著提高受损神经元的细胞活力(MTT值),降低培养液中的乳酸脱氢酶含量。结论广枣水提液及其小分子组分对拟衰老神经元有显著的直接和间接保护效果,其作用机制可能与包括广枣总黄酮在内的小分子组分抗氧化,抑制细胞钙超载,刺激星型胶质细胞分泌生物活性物质有关。

Abstract:Purpose To study the effects and the possible mechanisms of Choerospondias axillaries fruit(CAF) on rat cultured cortex neuron injury induced by Serum-Free DMEM.MethodsTo observe the effects of CAF and its macro-molecular part and its micro-molecular part on rat cultured cortex neuron injury induced by Serum-Free DMEM and effects of astrocytes′ culture stimulated by CAF and its micro-molecular part and its macro-molecular part on rat cultured cortex neuron injury induced by Serum-Free DMEM.ResultsCAF and its micro-molecular part and Astrocytes′ culture stimulated by CAF and its micro-molecular part can significantly improve the activity of injuried neuron detected by MTT method and decrease the extraction of lactatede hydrogenase(LDH) released by injuried neuron.ConclusionCAF and its micro-molecular part can significantly protect injuried neuron directly or by astrocytes.The possible mechnism is related to the activities of its micro-molecular part including total flaconoids of fructus chorspondiatis(TFC),which can prevent oxidation and inhibit Ca2+ overload and stimulate astrocyte to excrete bio-active factors.
