多巴胺受体D2基因TaqI位点多态性与高血压病的关系

摘    要:目的 探讨多巴胺受体D2基因TaqI位点多态性与原发性高血压病的相关性。方法 运用聚合酶链反应一限制性片段长度多态性法(PCR-RFLP)分析TaqI位点A1A1,A1A2,A2A2基因型在原发性高血压病组和对照组的分布情况。结果 等位基因A1,A2在原发性高血压病组和对照组的分布频率分别为0.34,0.66和0.43,0.57。基因频率分布符合Hardy-Weinberg平衡,样本具有群体代表性,两组人群的基因型和等位基因频率比较,差异有显著性(P<0.05)。原发性高血压病组中A2等位基因舒张压明显高于A1等位基因舒张压(P<0.05)。结论 在湖北省人群中,多巴胺受体D2基因TaqI位点多态性与原发性高血压病显著相关。

Abstract:Objective To explore the relationship between dopamine D 2 receptor gene TaqI polymorphism and primary hypertension in Chinese population in Hubei province.Methods The distribution of A 1A 1,A 1A 2,A 2A 2 genotypes of TaqI polymorphism between primary hypertensive patients and control group were identified by using polymerase chain reaction based restriction fragment length polymorphism.Results The allelic frequencies were 0.34 for the A 1 allele and 0.66 for the A 2 allele in primary hypertension patients, 0.43 for the A 1 allele and 0.57 for the A 2 allele in control group. The allelic frequencies were in Hardy Weinberg equilibrium. Significant differences in the genotype distribution( P <0.05) and allelic frequencies( P <0.025) between hypertensive and control group were seen,and the diastolic blood pressure for A 2 allele was higher significantly than that for A 1 allele in primary hypertensive subjects( P <0.05).Conclusions The TaqI polymorphism of human dopamine D 2 receptor relates significantly with primary hypertension in Chinese population in Hubei province.
