脑出血24h后不同蛋白酶激活受体表达的初步筛查

摘    要:目的初步探讨脑出血后血肿周围组织的不同蛋白酶激活受体(PARs)的表达情况。方法Wistar大鼠20只,随机分为脑出血组(采用2次注血/2次退针法建立脑出血大鼠模型,右侧尾状核注入自体血50μL)和对照组(右侧尾状核注人生理盐水50μL)。术后24h将大鼠处死取脑,制作冰冻切片,采用免疫荧光染色(间接法)检测血肿周围组织PAR-1、PAR-2、PAR-3、PAR-4阳性细胞的表达情况。结果①脑出血组,出血后24h血肿周围组织PAR-1表达明显增多,与对照组比较P〈0.01。②恼出血组,出血后24hPAR-2主要在血管内皮细胞表达,与对照组比较无明显差异。③脑出血组,出血后24h无PAR-3、PAR-4荧光表达,与对照组比较无明显差异。结论脑出血24h后血肿周围组织PAR-l表达增加。

Preliminary Screening on Protease Activated Receptors in Perihematoma in a Rat Model of IntracerebralHemorrhage

Abstract:Aim To probe the expression of different protease activated receptors (PARs) in perihematoma in a rat intracerebral hemorrhage (ICH) model preliminarily. Methods 20 rats were divided randomly into two groups: sham and ICH groups. An ICH model of rat was established by double injection and double withdrawal. All rats were killed to take brains after the models established 24 h, and frozen sections of brain were acquired. The positive cells of PAR-1/PAR-2/PAR-3/PAR-4 were probed by immunofluorescence. Results ①The expression of PAR- 1 positive cells in perihematoma in ICH group was more than that in the sham group(P〈0.01). ②The immunofluorescence of PAR-2 was found in capillary vessel like tissue. There was no significant difference between the ICH group and the sham group. ③ No obvious immunofluorescence of PAR-3 or PAR-4 was probed in ICH model, so was in the sham. Conclusion The major thrombin receptor up-regulated in perihematoma 24 h after ICH in a rat model was PAR-1.
