P27kipl和cyclin E在肾细胞癌中表达的研究

目的 :探讨细胞周期素依赖激酶抑制蛋白P2 7kipl(P2 7)和细胞周期素E(cyclinE)在肾癌中表达情况 ,并研究二者与肾癌生物学行为的关系。方法 :采用免疫组化SP法测定 4 2例肾透明细胞癌及 2 0例癌旁正常肾组织石蜡切片中P2 7蛋白和cyclinE蛋白的表达。结果 :P2 7在癌旁正常肾组织中的阳性率为 2 0 .0 % ,在肾细胞癌组织中的阳性率为 5 2 .4 % (P <0 .0 1)。P2 7在肾癌组织分级G1、G2 、G3 的阳性率分别为 83.3%、6 8.7%、30 .0 % ,P2 7表达与肾癌的分级呈负相关 (P <0 .0 1)。CyclinE在癌旁正常肾组织中的阳性率为 30 .0 % ,在肾细胞癌组织中的阳性率为 5 4 .8% (P <0 .0 1)。CyclinE在G1、G2 、G3 ,肾癌组织中的阳性率分别为 83.3%、6 8.8%、35 .0 % ,cyclinE蛋白的表达与肾癌的分级亦呈负相关 (P <0 .0 5 )。P2 7和cyclinE在肾癌中的表达呈正相关 (P <0 .0 1) ,r =0 .5 4。结论 :P2 7和cyclinE与肾癌的组织学分级呈负相关 ,提示 :P2 7和cyclinE有可能作为判别肾癌分化程度的标志物 ,并可能成为判断肾癌预后的有用指标。

Expression of P27kipl protein and cyclin E protein in renal cell carcinoma

Objective:To investigate the expressions of P27kipl (P27) protein and cyclin E protein in human renal cell carcinoma and in paracarcinoma kidney cortex tissue,and to explore the correlation of these protein expressions with biological behavior of tumors.Methods:P27 and cyclin E expressions were studied in formalin-fixed,paraffin-embedded sections of renal cell carcinoma and in paracarcinoma kidney cortex tissue by means of a specific and sensitive immunohistochemical S-P assay.Results:The positive rates of P27 protein and cyclin E protein in renal cell carcinoma were significantly higher than those in paracarcinoma kidney cortex tissues ( P <0.01),and P27 and cyclin E were concurrently expressed in RCC.There was an inverse relationship between the expression rate of P27 and the histological tumor grades ( P <0.01),and the same with cyclin E.Conclusion:P27 and cyclin E expression rates are negatively correlated with the malignant degree of RCC,indicating that P27 and cyclin E might be a useful prognostic marker for renal cell carcinoma.
