白细胞介素-17在阿霉素肾病大鼠肾小球硬化中的作用

摘    要:目的 观察白细胞介素-17(interleukin-17,IL-17)在阿霉素肾病大鼠肾组织中的表达,并探讨其在阿霉素肾病大鼠肾小球硬化中的作用.方法 雄性SD大鼠分为正常对照组和阿霉素肾病组(模型组),每组8只,于第2、4、8、12周分别处死.光镜观察肾小球的病理学情况并分析肾小球硬化指数;采用半定量RT-PCR法检测肾组织IL-17 mRNA的表达;ELISA法检测肾组织匀浆上清中IL-17和IL-1β蛋白水平;免疫组化观察IL-17在肾脏的定位表达.结果 第8周模型组肾组织IL-17 mRNA水平、IL-17和IL-1β蛋白水平均高于正常组(P=0.041、P=0.000、P=0.007);第12周均进一步升高(均P=0.000),并且IL-17与IL-1β、肾小球硬化指数均呈明显的正相关(P=0.037、P=0.021).结论 IL-17在阿霉素肾病大鼠硬化的肾组织中表达明显增加,可能在阿霉素肾病大鼠发生肾小球硬化过程中起了一定的作用.

Abstract:Objective To investigate the role of interleukin-17 (IL-17) in glomerulosclerosis secondary to adriamycin-induced nephropathy in rats. Methods Male Sprague-Dawley rats were divided into normal control group and adriarnycin-induced nephropathy group(model group). Eight rats were sacrificed at 2nd week, 4th week, 8th week, 12th week respectively. Histopathological changes of renal tissue were observed under light microscope and glomerulosclerosis index was evaluated. The expression of IL-17 was measured by RT-PCR, ELISA and immunohistochemistry. The expression of IL-1β was detected by ELISA. Results Compared with control group, the expressions of IL-17 mRNA, IL-17 and IL-1β protein were higher in model group of the 8th week (P=0.041, P=0.000 and P=0.007 respectively), even they were further higher in model group of the 12th week than that in the control group (all P=0.000). In model group of the 12th week, the level of IL-17 protein had positive correlation with IL-1β and glomerulosclerosis index (P=0.037, P=0.021 respectively). Conclusion IL-17 expression is elevated and may contribute to glomerulosclerosis in adriamycin-induced nephropathy.
