摘    要:目的:探讨药物依赖行为和酪氨酸羟化酶在成瘾过程中的相互关系.方法:建立大鼠条件性地点偏好模型,使用免疫印迹法在整个成瘾过程中对腹侧被盖区和伏隔核内的酪氨酸羟化酶免疫反应性进行监测.结果:大鼠地点偏好和腹侧被盖区酪氨酸羟化酶水平均在实验第3日发生剧烈变化,之后表现出相似的变化趋势.结论:动物成瘾行为与酪氨酸羟化酶的变化密切相关,酪氨酸羟化酶的调节可能是吗啡成瘾的分子基础.

Synchronized changes of TH immnoreactivity in VTA and conditioned place preference by chronic morphine in rats
Abstract:AIM: To observe the relationship between the drug dependence behavior and levels of tyrosine hydroxylase (TH) in drug acquisition process. METHODS: Male SD rats were used and psychological dependence was established by chronic morphine administration and detected by conditioned place preference (CPP). The TH immuonreactivity in ventral tagment area (VTA) and nuclear accumbens (NAC) was monitored by immunobloting. RESULTS : The CPP and TH in VTA of rats began to increase rapidly at the 3rd day and kept this increasing trend up to the 7th day( P <0.01). CONCLUSION: There is a close correlation between the drug dependence behavior and the TH regulation. The regulation of TH in mesolimbic system may be responsible for the drug addiction.
