褪黑素对重症急性胰腺炎肺损伤的保护作用

摘    要:目的 :观察褪黑素 ( MLT)对重症急性胰腺炎 ( SAP)时肺组织脂质过氧化损伤的保护作用。方法 :采用胆胰管末端穿刺逆行注入 5 %去氧胆酸钠 ( 1 ml·kg-1体重 ) ,制成 SAP动物模型 ,于皮下注入 MLT溶液或相同体积的生理盐水 ( NS) ,术后 6h及 1 2 h活杀大鼠检测肺组织丙二醛( MDA)含量、髓过氧化物酶 ( MPO)活性、超氧化物歧化酶 ( SOD)活性及谷胱甘肽过氧化物酶( GSH- PX)活性。结果 :SAP+ MLT组大鼠肺组织炎性病理改变较 SAP+ NS组明显减轻 ,MDA含量及 MPO活性均较 SAP+ NS组显著降低 ( P<0 .0 5 ) ,肺组织 SOD及 GSH- PX活性较 SAP+NS组显著升高 ( P<0 .0 5 )。结论 :氧自由基所引发脂质过氧化损伤是 SAP肺损害的重要因素 ,MLT可有效减轻肺组织脂质过氧化损伤

Abstract:Objective:To investigate the protective effect of melatonin (MLT) on the lung lipid peroxidization injury in severe acute pancreatitis. Methods:Severe acute pancreatitis (SAP) model was induced by retrograde injection of 5% sodium deoxycholate into the biliopancreatic duct in a volume of 1 ml·kg -1 body weight in rates. Saline solution containing MLT (50 mg·kg 1 body wt) in the SAP+MLT group was injected subcutaneously whereas only the same volume of saline solution was injected in SAP+NS group. Animals were killed at the 6th and 12th hour,respectively, after operation. Lung malondialdehyde (MDA) content, superoxide dismutase (SOD) activity, glutathine peroxidase (GSH Px) activity and myeloperoxidase (MPO) activity were determined. Results: Histopathologically, the imflammatory lesions of the lung in SAP+MLT growp were much wilder than those in SAP+NS group, compared with SAP+NS group, lung MDA content and MPO activity at the 12th hour after the operation were decreased dramatically in SAP+MLT group, but SOD activity and GSH Px activity were increased significantly ( P <0.05 for both). Conclusion:Free radicals may play an important role in the pathogenesis of lung lipid peroxidization injury in severe acute pancreatitis and MLT can reduce the severity of the injury.
