肺腺癌相关蛋白的筛选及鉴定

目的建立肺腺癌组织及配对癌旁组织的2-DE图谱,分析两者蛋白质表达的差异,进而寻找有诊断价值的相关蛋白。方法应用双向凝胶电泳(2-DE)和基质辅助激光解吸飞行时间质谱(MALDI-TOF-MS)对肺腺癌组织、配对癌旁组织进行蛋白质组学比较研究。结果获得了分辨率高、重复性好的双向凝胶电泳图谱。经过Im-ageMaster 2DPlatinum 5.0软件对图像分析后,获得差异大于3倍的蛋白点63个,12个点仅在肺腺癌组织中表达,5个点仅在配对癌旁组织中表达。质谱鉴定去冗余后确定了5种蛋白质,其中4种蛋白与肿瘤相关,分别为S100钙结合蛋白(S100-A11)、Ras相关蛋白Rab-14、微管辅助蛋白(Stathmin)、凝溶胶蛋白(gelsolin)。结论肺腺癌组织与配对癌旁组织的双向电泳结果有明显差异,这些差异蛋白可作为肺腺癌的候选肿瘤标志物。

Screening and Identification of Lung Adenocarcinoma Associated Proteins

Objective To establish the two-dimensional electrophoresis patterns of the protein in human Lung adenocarcinoma and paired cancer tissues,and to analyze the differences in proteins expression,then to screen associated proteins for diagnosis.Methods Two-dimensional gel electrophoresis(2-DE) and matrix-assisted laser desorption/ionization time of flight mass spectrometry(MALD I-TOF-MS)were employed to study proteomics between the lung adenocarcinoma and paired cancer tissues comparatively.Results Two-dimensional electrophoresis patterns of tissues proteins with high-resolution and reproducibility were obtained.Sixty-three significantly differentially expressed protein spots were screened with ImageMaster 2D Platinum 5.0 software,and twelve protein spots were expressed only in the lung adenocarcinoma,and five protein spots were expressed only in the paired cancer tissues.Five proteins were successfully identified via peptide mass print using MALDI-TOF-MS,among which four proteins were reported to be related to lung,such as S100-A11,Ras related protein Rab-14,Stathmin and gelsolin.Conclusion There are significant differences between the lung adenocarcinoma and paired cancer tissues,which may be special tumor markers of the lung adenocarcinoma.
