醋酸铅对人股动脉内皮细胞和星形胶质细胞的毒性效应

摘    要:目的观察醋酸铅对动脉内皮细胞和星形胶质细胞的毒性效应,探讨醋酸铅通过血脑屏障的可能机制。方法醋酸铅处理动脉内皮细胞和星形胶质细胞后进行Giemsa或HE染色,检测观察细胞形态学变化;细胞免疫化学法检测P53、Bax、Bc1-2的表达,流式细胞仪检测细胞凋亡率。结果经10μmol/L醋酸铅处理24h后Giemsa,H.E染色均显示出醋酸铅组细胞成凋亡形态学改变。动脉内皮细胞和星形胶质细胞的细胞免疫化学结果都显示醋酸铅组的P53和Bax阳性表达升高,而Bc1-2的阳性表达下降。其流式细胞仪检测结果表明醋酸铅组细胞凋亡率较对照组明显升高。结论醋酸铅可损伤动脉内皮细胞和星形胶质细胞,促进凋亡及影响血脑屏障。

Abstract:Objective]To observe the toxic effect of lead acetate on human femoral artery endothelial cells (HFAEC) and astrocytes,and explore the mechanism of lead acetate passing through blood-brain barrier.Methods]After treated with lead acetate,Giemsa and H.E.staining was applied to observe the morphological changes of these 2 cells,and the expression level of P53,Bax,Bc1-2 were detected with immunohistochemical method.The rate of apoptosis was measured by flow cytometry.Results]After treated with 10 μmol/L lead acetate for 24 hours,Giemsa and H.E.staining showed that morphological changes of apoptosis cells were observed in the lead acetate group.Immunohistochemical analysis indicated that the positive expression of P53 and Bax increased while the expression of Bc1-2 decreased.The flow cytometry results showed the higher cell apoptosis rate in the treated group than that in the control group.Conclusion]Lead acetate can damage HFAEC and astrocytes,promote apoptosis,and affect the blood-brain barrier.
