Wt1反义寡核苷酸阻断白血病微小残留病wt1基因表达的体外研究

摘    要:本研究通过wt1反义寡核苷酸(ASO)抑制wt1基因的表达,观察微小残留病模型及患者基因表达的变化。取56例复诊急性白血病完全缓解(CR)患者骨髓,用RT-PCR方法筛选出wt1基因表达阳性的骨髓标本。白血病细胞系微小残留病(MRD)模型及筛选出的wt1阳性急性白血病CR患者骨髓标本进行了细胞培养并用wt1ASO处理细胞,观察wt1基因表达的变化。结果表明:wt1基因的敏感性是10-3-10-4;56例急性白血病CR患者骨髓wt1基因的表达阳性率为16%。用wt1ASO处理wt1表达阳性的K562、HL-60细胞系MRD模型以及9例存在MRD的急性白血病CR患者骨髓,发现wt1ASO组有效的阻断了wt1基因的表达,而正义寡核苷酸组和对照组wt1基因的表达仍为阳性。结论:wt1ASO体外能够阻断白血病MRD中残留白血病细胞wt1基因的表达。
