| 摘 要： | 目的应用高效液相色谱法(HPLC)测定大鼠血浆中精氨酸右布洛芬浓度,研究单次口服给药后在大鼠体内的药代动力学。方法 SD大鼠分别按照体质量单次口服精氨酸右布洛芬50 mg/kg后,采用HPLC测定血浆中精氨酸右布洛芬药物的浓度,绘制血药浓度-时间曲线,计算其药代动力学参数。结果单次口服精氨酸右布洛芬药-时曲线均符合口服吸收的一级动力学二室模型,主要药代动力学参数:t1/2α为(1.956±1.136)h,t1/2β为(5.551±3.981)h,Cmax为(7.365±2.627)μg/L,AUC(0-12)为(18.807±4.007)μg/(L.h),AUC(0-∞)为(23.852±7.767)μg/(L.h)。结论 HPLC法能够准确地测定精氨酸右布洛芬血药浓度,能满足药代动力学的研究需求。
