亚硒酸钠对SGC-7901细胞增殖的影响

摘    要:目的:研究亚硒酸钠对SGC-7901增殖的影响.方法: 应用集落形成试验研究了亚硒酸钠对SGC-7901细胞集落形成的影响;应用流式细胞仪检测了亚硒酸钠对SGC-7901细胞生长周期的影响;应用电镜及流式细胞仪观察了亚硒酸钠诱导SGC-7901细胞凋亡的作用.结果: 亚硒酸钠对SGC-7901细胞的集落形成有明显的抑制作用,其抑制作用与其作用浓度和作用时间呈正相关.流式细胞仪检测显示亚硒酸钠作用SGC-7901细胞24 h后,细胞周期发生改变,G1期细胞百分率减少,S期细胞百分率增加,DNA直方图上出现典型的亚二倍体细胞的“凋亡峰”;电镜下可见细胞核固缩,染色质凝集呈新月形紧贴于核膜周边,核膜扭曲等特征.结论: 亚硒酸钠对SGC-7901细胞的增殖具有抑制作用,抑制作用程度与作用浓度和时间呈正相关;亚硒酸钠抑制SGC-7901细胞生长的机理之一是诱导细胞凋亡.

Abstract:Objective: To examine the effect of sodium selenite on growth of SGC-7901 cells.Methods: The effect of sodium selenite on SGC-7901 cell colony-forming was examined by using colony-forming test;the effect of sodium selenite on SGC-7901 cell growth cycle was measured by using flow cytometry;sodium selenite-induced SGC-7901cell apoptosis was examined by electron microscope and flow cytometry.Results: Sodium selenite had a significant inhibitory effect on SGC-7901 cell colony-forming with dose-effect relationship.Flow cytometry detection showed that,after 24 hours,sodium selenite reduced the percentage of G_1 phase cells and increased the percentage of S phase cells in cell cycle.DNA histogram showed a typical apoptosis peak as hypodiplod.Electron microscope detection showed nucleus pycnosis,crescent-shaped chromatic agglutination closed to karyotheca and karyotheca distortion.Conclusion: Sodium selenite has an inhibitory effect on SGC-7901 cell growth with dose-effect relationship.Induction of cell apoptosis may underlie the mechanism of the inhibitory effect of sodium selenite on SGC-7901 cell growth.
