肺纤维化动物模型肺组织形态定量分析

摘    要:目的:采用光镜组织病理学检查及电子计算机图像分析仪形态定量分析,更客观地评价肺纤维化形成的过程。方法:气管内一次注入博莱霉素A5(BLMA5)制成肺纤维化动物模型,于第7、14、28、43、56天分批处死动物,光镜下依据Szapiel等的方法测定肺泡炎和肺纤维化程度,同时采用电子计算机图像分析仪测算肺泡腔、肺泡炎及肺纤维化所占面积。结果:注入BLMA5第7天肺泡炎最重,第28天肺纤维化最明显。结论:组织学定量与电子计算机图像分析仪所测得结果有良好的相关性。采用本方法可以客观地评价各种药物治疗肺纤维化的疗效。

Abstract:To evaluate objectively the formation process of pulmonary fibrosis.METHODS:Animal model of pulmonary fibrosis was established by injection of bleomycin A5. The status of pulmonary alveolitis and pulmonary fibrosis was measured microspically according to S.V. Szapiel method on the samples that were taken on day 7, 14, 28, 42 and 56 after injection,and the area of al veolar cavity, alveolitis and pulmonary fibrosis with computer imaging analysis.RESULTS: A serious pulmonary alveolitis was found on day 7 and obvious fibrosis on day 28 after injection. CONCLUSIONS: The results obtained with histologically qualitative analysis is well correlated to that obtained with computer imaging analysis. This method is helpful in evaluating treatment effect of various kinds of medicine on pulmonary fibrosis.
