PCR动力学数学模型在PCR产物直接核酸序列分析中的应用

摘    要:使用PCR动力学数学模型提供的PCR产物数量计算方法计算经PCR扩增获得的产物数量,纯化后用於PCR产物的核酸序列分析。由於PCR动力学数学模型提供了比较准确的产物数量计算,使测序模扳数量总能保持在比较适当的范围,因此测序结果的可读性、自显影条带的清晰度保持了较好的水平。提示了PCR动力学数学模型是基本正确的。说明准确的定量计算PCR产物数量对PCR产物直接核酸序列分析是有益的。

Abstract:To apply the mathematical model of PCR kinetics,the quantity of the PCR product was calculated and ,after pured,its nucleic acid sequence was analysised directly.As the model provided a exact method of calculation of the PCR product quantity,the amount of sequenced templates may reach to a suitabl level. The autoradiogrphy of sequencing results was readable and its visibility kept rather satisfaction,the results suggested that the PCR kinetic mathematical model is accurate to estimate the quantity of the PCR product.the correct quantitation of templates is benefit for direct nucleal acid sequencing.
