孕期小鼠巨细胞病毒胎盘感染对单核-巨噬细胞胞毒作用的影响

摘    要:目的:应用孕期小鼠巨细胞病毒(MCMV)感染模型,观察MCMV感染后孕鼠腹腔单核-巨噬细胞胞毒作用。方法:用ELISA法筛选无MCMV感染史、性成熟期BALB/c小鼠,雌雄同笼受孕。孕龄12.5天时,模型组胎盘显微注射MCMV,对照组注射细胞维持液,在孕龄18.5天处死母鼠,以MTT比色法检测小鼠腹腔液中单核-巨噬细胞细胞毒性百分率。结果:细胞毒性百分率对照组为(96.2±2.2)%,模型组为(19.3±17.4)%,两组差异具有极显著性意义(P<0.01)。结论:孕期胎盘MCMV感染明显降低孕鼠腹腔液中单核-巨噬细胞胞毒作用。

Abstract:Objective:Study the monocaryon---macrophage toxic action after intrauterine murine cytomegalovirus(MCMV)infection.Methods:The mature BALB/c mice without MCMV infection identified by Enzyme-linked immunoabsorbent assay(ELISA)were served as the subjects.We put the male and the female in the same cages,and then we injected MCMV into the placentas of the model group mice at day 12.5 of gestation,the control group were injected with cell maintenance medium,executed female mice at day 18.5 of gestation;observed the cytotoxicity percentage of the monocaryon-macrophage in peritoneal fluid by MTT Assay.Results:The cytotoxicity percentage of the mold group was(96.2±2.2)percentage,the control group was(19.3±17.4)percentage,the difference between two groups was significant(P<0.01).Conclusion:The monocaryon-macrophage toxic action in peritoneal fluid was down obviously in duration of pregnancy placental infection by MCMV.
