蛇床子素在正常与急性肾功能衰竭兔体内的分布研究

目的:正常与急性肾功能衰竭兔iV给药后,比较研究蛇床子素在兔体内的动态定量分布特点.方法:蛇床子素iv给药后,分时采集血样,断头处死动物,取出心脏、肝脏、肾脏、脑组织并制备匀浆,以HPLC法测定血浆及组织中蛇床子素的含量.结果:iv给药后,以同一器官不同时间药物的平均含量占总量百分比的多少表示,蛇床子素在正常兔体内的分布依次为血液＞心脏＞肾脏＞脑＞肝脏.而在肾衰兔体内的分布为血液＞肾脏＞心脏＞脑＞肝脏,与正常组相比,蛇床子素在肾衰兔体内血液、肾脏中含量显著升高(P＜0.05),而在心脏、脑、肝脏组织的含量显著降低(P＜0.05).结论:蛇床子素在正常与肾衰兔体内组织中的含量分布有明显区别.

Distribution in vivo of osthole in normal and renal failure rabbits

AIM: To compare the distribution pattern of osthole in normal and renal failure rabbits after i.v. administration. METHODS: After i.v. administration of osthole, rabbits were executed at different time. The blood samples and homogenates of heart, liver, kidney and brain were prepared at various intervals. The levels of total osthole in blood and the organ homogenates were determined by HPLC, respectively. RESULTS: The distribution of osthole in normal rabbits was found in blood>heart>kidney>brain>liver after i.v. administration. The distribution of osthole in renal failure rabbits was found in blood>kidney>heart>brain>liver after i.v. administration. The distribution of osthole in blood and kidney were siginificantly higher( P <0.05); in heart, brain and liver were siginificantly lower( P <0.05) in renal failure groups than normal groups. CONCLUSION: There is siginificant difference in the distribution of osthole between the normal and the renal failure rabbits.
