鸡胚脊髓运动神经元对神经生长因子的应答及其钙依赖性研究

摘    要:观察神经生长因子(nervegrowthfactor)在钙拮抗剂和不同CJ“浓度条件下,对鸡胚脊髓运动神经元生长发育的影响。方法:取3天胚龄的鸡胚获取脊髓运动神经元。实验分组:对照组,NGF组,NGF+Verapamil组和NGF+Ca2+组。观察项目:神经元最大突起平均长度,运动神经元计数。运动神经元百分比和胆碱酯酶染色。结果:培养1周,NGF组运动神经元数及百分比和突起长度明显超过对照组和NGF+Verapamil组(P<0.01)。Ca2+浓度4mM组的突起长度明显大于其它Ca2+浓度组(P<0.01)。结论:NGF对鸡胚脊髓运动神经元的生长、发育有促进作用,且明显依赖于跨膜Ca2+内流。

Abstract:To establish a method for culture of motorneurons from spinal cords of chicken embryos and to observe the responds of motoneurons to nerve growth factor(NGF) and the influence of calcium . 3 days chick- en embryos were used to obtain the znotoneurons of ventral spinal cords . Four experimental groups were as fol - Iows: control group,NGF group, NGF veraparmil group and NGF Ca2 grop . The mean length of the longest neurits of motoneurons, number of motoneuruns, percentage of motoneurons were cdculated and cholinesterase histochemical and electron microscopical studies were done . The results showed that the mean of longest neuritetes, number and percentage of motoneurons in NGF group surpassed sigmtificantly those in control grop and NGF . verapamil group at 7 days of culture, and the mean length of longest neurite was longer in 4.M Cal group than in other Ca2 growp. lt was concluded that NGF could promote development and gmwth of motoneurons from embryonic chicken spinal cords and it depended on influx of extracellular calcium Into rlcurons .
